Abstract The aim of the present study was to evaluate semen cryopreservation with ACP-Lact® diluent, which consists of coconut water powder (ACP) added to goat milk powder. After thawing, the samples were evaluated for sperm kinetics, membrane evaluation and in vivo insemination. For cryopreservation, a pool was made with the ejaculate of six goats, diluted in four equal aliquots for the respective treatments: T1 (ACP-Lact®); T2 (ACP-Lact® 50%); T3 (ACP + 2.5% egg yolk) and T4 (Tris + 2.5% egg yolk). After dilution of the treatments, the samples were placed in 0.5 ml straws and chilled at a rate of -1.07°C/min. After reaching 4°C and stabilizing for one hour, the straws were placed in nitrogen vapour at -60°C for 15 minutes and then immersed in liquid nitrogen (-196ºC). The straws were thawed in a 37°C water bath and kinetic assessments were performed immediately using a computerized semen analysis program (CSA), viability (EN), membrane functionality (HOST), mitochondrial activity (DAB) and DNA integrity assessment of spermatozoa. For the in vivo experiment, ten goats were inseminated, divided into two groups of five goats each, G1 inseminated with ACP-Lact® and G2 with ACP, by fixed-time artificial insemination (FTAI). Regarding the kinetic parameters, the ACP-Lact® treatment showed higher progressive motility (PM) and sperm velocity than the other treatments (36.77%). In the VSL parameter the ACP-Lact diluent was superior to ACP and Tris. In viability the treatment with ACP-Lact® was superior to the treatment with Tris, 95% and 83% respectively. In FTAI two goats were born out of the 5 goats inseminated with ACP-Lact®. It was concluded that the use of ACP-Lact® for cryopreservation of caprine semen is efficient in maintaining seminal parameters during thawing in vitro and in vivo and proved to be a good alternative extender for the caprine species.

Abstract Effects were assessed of the dilutants TRIS and ACP - 101c® with the addition of different guinea fowl (Numida meleagris) egg yolk concentrations. Fifteen ejaculates were collected from five goats of the Anglo Nubian breed. The ejaculates were pooled and then divided into 12 groups, two control groups (GC1 TRIS, with 2.5% Gallus gallus domesticus hen egg yolk GOGD), (GC2 Control Group ACP - 101c®, with the addition of 2.5% Gallus gallus domesticus hen egg yolk GOGD) and ten experimental groups (EG), containing TRIS and ACP added with different concentrations of egg yolk from guinea hen (Numida meleagris) (TRIS 2,5% GONM; TRIS 5% GONM; TRIS 10% GONM; TRIS 15% GONM; TRIS 20% GONM; ACP® 2,5% GONM; ACP® 5% GONM; ACP® 10% GONM; ACP® 15% GONM; ACP® 20% GONM). Then cryopreservation was carried out and the samples stored in liquid nitrogen (-196 °C). After seven days, the samples were thawed and assessed for spermatic kinetics, immunofluorescence and sperm morphology. Analysis of GOMN by the CASA system showed that the various parameters were similar to those of GOGD (P>0.05). The membrane integrity, mitochondrial potential and the acrosome were not influenced by the treatment (P>0.05) nor by the dilutant used for cryopreservation (P>0.05). The spermatic morphology was also preserved by the different GOGD and GONM concentrations in the ACP® and TRIS dilutants, with no statistically significant differences (P<0.05). It was concluded that Numida meleagris egg yolk, as external membrane cryoproctant added to the dilutants ACP-101c® and TRIS, improved goat semen quality.

ABSTRACT Toxoplasmosis is a zoonosis of reproductive nature that is transmitted to humans mainly by ingestion of undercooked meat or drinking oocyte-contaminated water. The slaughter of small ruminants in clandestine slaughterhouses and the presence of companion animals in these locations may be epidemiologically important for the dissemination of the disease. The objective of the present study was to determine toxoplasmosis prevalence by researching anti- Toxoplasma gondii antibodies in meat sheep herds from farms in Teresina microregion, Piauí, Brazil. A total of 450 blood samples were collected from sheep of both sexes, belonging to 28 herds from the 14 municipalities that comprise the microregion. The samples were analyzed by the enzyme-linked immunosorbent assay (ELISA), and the presence of anti-T. gondii antibodies was observed in 62% (279/450) of the animals in all the municipalities and on at least one farm in each municipality. The high occurrence of positive animals is an indication of reproductive problems in these herds, characterizing a problem for both sheep rearing and public health.

RESUMO: Dentre as enfermidades que acometem o sistema reprodutivo dos animais domésticos, a brucelose na espécie ovina tem se destacado por gerar prejuízos econômicos significativos à ovinocultura. Dessa forma, apresenta-se como uma ameaça ao crescimento e à produtividade dos rebanhos ovinos. Diante de tal problemática, objetivou-se, por meio desta pesquisa, identificar a prevalência de brucelose ovina em rebanhos pertencentes a municípios da microrregião de Teresina, Piauí, por meio dos testes sorológicos, imunodifusão em gel de ágar (IDGA) e ensaio imunoenzimático (ELISA) indireto. Quatorze municípios foram incluídos na pesquisa. Para sua execução, colheram-se, por punção venosa da jugular, amostras sanguíneas de 521 animais púberes. Após colheita, as amostras foram submetidas às técnicas sorológicas, IDGA e ELISA indireto, para a detecção de anticorpos anti-B. ovis. Das 521 amostras submetidas ao teste de IDGA, 23 (4,41%) foram sororreagentes e 498 (95,58%) negativas. Quanto ao teste ELISA indireto, obtiveram-se 24 (4,61%) amostras suspeitas e 497 (95,39%) amostras negativas, não havendo animais reagentes neste teste, apenas suspeitos. A soroprevalência da brucelose ovina na microrregião homogênea de Teresina, Piauí, foi de 4,41%. Assim, foi possível identificar ovinos com sorologia reagente à infecção por B. ovis, presente em municípios do estado do Piauí. Além disso, a IDGA mostrou-se mais sensível em detectar animais que tiveram contato com o agente etiológico da doença.

ABSTRACT: Among the diseases that affect the reproductive system of domestic animals, brucellosis in the sheep species is important because it generates significant economic losses to sheep rearing. Thus, it is a threat to the growth and productivity of sheep herds. In the face of this problem, the objective of the present research was to identify the prevalence of ovine brucellosis in herds in municipalities of the Teresina, Piauí, Brazil microregion by using the agar gel immunodiffusion assay (AGID) and indirect enzyme-linked immunosorbent assay (ELISA) serological tests. Fourteen municipalities were included in the research. Blood samples were collected from 521 pubescent animals by puncturing the jugular vein. After collection, the samples were submitted to the serological techniques, AGID and indirect ELISA, to detect anti B. ovis antibody. Of the 521 samples submitted to the AGIDtest, 23 (4.41%) were sera reagent and 498 (95.58%) negative. The indirect ELISA tests, 24 (4.61%) suspect samples and 497 (95.39%) negative samples were obtained, and there were no reagent animals in this test, only suspect. The seroprevalence of ovine brucellosis in the Teresina, Piauí microregion was 4.41%. Thus, it is possible to identify sheep with reagent serology to infection by B. ovis, that is present in municipalities in the state of Piauí, Brazil. Furthermore, AGIDwas shown to be more sensitive in detecting animals that had had contact with the etiological agent of the disease.

RESUMO: Lentivírus de pequenos ruminantes (LV) é o termo genérico utilizado para designar os vírus da artrite encefalite caprina e Maedi-Visna, os quais pertencem à família Retroviridae, subfamília Orthoretrovirinae, gênero Lentivirus. Tais vírus infectam caprinos e ovinos, causando enfermidades de curso lento com lesões inflamatórias, crônicas e degenerativas que podem atingir vários órgãos, provocando caquexia e morte. Os animais infectados eliminam o vírus sobretudo por meio de secreções e excreções e transmitem-no especialmente em situações de estreito contato. Não há tratamento até o momento. O controle é baseado na criação segregada, no manejo e no sacrifício dos positivos. Esse agente infeccioso já foi relatado em várias partes do mundo, sendo responsável por perdas econômicas significativas. Por o agente ter sido verificado em vários estados do Brasil e por não existirem dados soroepidemiológicos nas mesorregiões sudeste e sudoeste piauiense, esta pesquisa teve por objetivo realizar inquérito sorológico para investigar a ocorrência de anticorpos para o LV em ovinos e caprinos nas microrregiões do Alto Médio Canindé, Picos e Floriano, no Piauí. Para tanto, foram coletadas 1.280 e 1.360 amostras de soro caprino e ovino, respectivamente, oriundos de 20 municípios, distribuídos nas três microrregiões, sendo o número de amostras proporcional ao rebanho efetivo de cada município. As amostras de soro foram analisadas utilizando o teste de imunodifusão em gel de agarose (IDGA). Nenhum dos soros pesquisados reagiu positivamente, constatando-se soroprevalência nula. Ressalta-se a importância da implantação de um rigoroso programa de controle para que se possa evitar a introdução e/ou a disseminação desse agente infeccioso nessas microrregiões.

ABSTRACT: Small ruminant lentiviruses (LV) is the generic term for the caprine arthritis-encephalitis and Maedi-Visna viruses, which belong to the Retroviridae family, Orthoretrovirinae subfamily, Lentivirus genus. The virus infects goats and sheep, causing slow course of disease with inflammatory, chronic and degenerative lesions, which can reach several organs, provoking cachexia and death. Infected animals eliminate the virus mainly through secretions and excretions and transmit it especially in close contact situations. There is no treatment to date. The control is based on segregated creation, management and sacrifice of the positive. This infectious agent has been reported in various parts of the world and is responsible for significant economic losses. It was verified in several states of Brazil and there are seroepidemiological data in southeast and southwest mesoregions of Piauí, Brazil. This research aimed to perform serological survey to investigate the occurrence of antibodies to LV in sheep and goats, in the regions of Alto Médio Canindé, Picos and Floriano. So, 1,280 and 1,360 serum goats and sheep samples, respectively, were collected, coming from 20 municipalities, distributed in the 3 microregions. The number of samples was proportional to the actual herd of each municipality. The samples were analyzed using the agar gel immunodiffusion test. None of the surveyed sera reacted positively, though there is a null seroprevalence. It was emphasized the importance of implementing a rigorous control program in order to prevent the introduction and spread of this infectious agent in these microregions.

Resumo: O objetivo deste trabalho foi avaliar os fatores de risco envolvidos na transmissão de Chlamydophila abortus a ovinos e caprinos, bem como verificar a soroprevalência da infecção. Foram selecionadas 110 propriedades, com um total de 500 amostras de ovinos - 350 da Microrregião do Alto Médio Gurgueia (MRAMG) e 150 da Microrregião Homogênea de Teresina (MRHT), ambas do Estado do Piauí - e 600 amostras de caprinos - 300 da MRAMG e 300 da MRHT. Um questionário epidemiológico foi empregado para identificar os principais fatores de risco. Os anticorpos anti-C. abortus foram detectados pela técnica de fixação de complemento. A prevalência geral da infecção por C. abortus foi de 7,2% (79/1.100). A prevalência em caprinos e ovinos na MRAMG foi de 7,9% (51/650) e, na MRHT, foi de 6,2% (28/450). Nas microrregiões estudadas, a prevalência em ovinos foi de 8,2% (41/500) e, em caprinos, de 6,3% (38/600). O sistema de criação, as práticas reprodutivas e o tipo racial foram fatores de risco relevantes para a infecção em ovinos. Também foram relevantes em caprinos as práticas reprodutivas e a origem das matrizes e dos reprodutores. A raça Dorper apresenta maior suscetibilidade à infecção por C. abortus.

Abstract: The objective of this work was to evaluate the risk factors involved in the transmission of Chlamydophila abortus to sheep and goats, as well as to verify the seroprevalence of the infection. One hundred ten farms were selected, with a total of 500 sheep samples - 350 from the Microregion of Alto Médio Gurgueia (MRAMG), and 150 from the Homogeneous Microregion of Teresina (MRHT), both in the state of Piauí, Brazil - and 600 goat samples - 300 of the MRAMG and 300 of the MRHT. An epidemiological questionnaire was used to identify the main risk factors. Anti-C. abortus antibodies were detected by the complement fixation technique. Overall prevalence of infection by C. abortus was 7.2% (79/1,100). The prevalence in goats and sheep in the MRAMG was 7.9% (51/650), and in the MRHT it was 6.2% (28/450). In the studied microregions, the prevalence in sheep was 8.2% (41/500), and in goats it was 6.3% (38/600). The raising system, reproductive practices, and racial type were significant risk factors for infection in sheep. Also significant in goats were the reproductive practices and the origin of the does and bucks. The Dorper breed shows increased susceptibility to infection with C. abortus.

As Lentiviroses de Pequenos Ruminantes (LVPR) incluem a Maedi-Visna (MV) em ovinos e a Artrite Encefalite Caprina (CAE). Essas enfermidades estão difundidas no mundo e são responsáveis por grandes perdas na produtividade destes animais. Os LVPR são vírus RNA da subfamília Lentivirinae que causam uma infecção persistente, sendo a detecção precoce uma das formas mais eficientes para limitar sua disseminação no rebanho. Visando contribuir com essas questões, este experimento foi realizado na Universidade Federal do Piauí (UFPI) em parceria com a Embrapa Caprinos e Ovinos, com o objetivo de padronizar a técnica de ensaio imunoenzimático indireto e compará-lo com a imunodifusão em gel de agarose no diagnóstico da CAE. Foram utilizadas 696 amostras de soros de caprinos machos e fêmeas oriundas do banco de soros da Unidade de Pesquisa de LVPR do Centro de Ciências Agrárias da UFPI. As amostras foram coletadas no período de janeiro de 2007 a março de 2010. Na padronização, verificou-se que 0,25 µg de proteína/poço, diluição de 1:200 do soro e concentração de 1:3.000 do conjugado anticorpo anti-IgG cabra apresentaram os melhores resultados. O ponto de corte obtido foi de 0,36. Na comparação, o Imunodifusão em Gel de Ágar (IDGA) detectou 128 (18,4%) amostras positivas, e o ELISA indireto (ELISA-i), 259 (37,2%). A sensibilidade e a especificidade do teste ELISA-i com relação ao IDGA foi de 94,5% e 75,7%, respectivamente. Verificou-se maior índice de positividade em caprinos acima de seis meses (p < 0,05), e nos machos obteve-se prevalência de 56,7% em comparação às fêmeas, 35,4%, (p < 0,01).

The Small Ruminant Lentiviruses (SRLVs) include Maedi-Visna (MV) of sheep and Caprine Arthritis-Encephalitis (CAE). These diseases are widespread and responsible for major production losses regarding sheep and goats. The SRLV is a RNA virus of the subfamily Lentivirus genus that causes persistent infections in goats. Early detection is one of the best ways to limit its spread in the herd. To contribute to these issues, this experiment was conducted at Universidade Federal doPiauí in partnership with Embrapa Goats and Sheep, with the objective of standardizing the technique of indirect ELISA (i-ELISA) and to compare it with Immunodiffusion in Agarose Gel to diagnose Caprine Lentiviruses (LC). Six hundred ninety six serum samples were used from the University Veterinary Hospital, Universidade Federal do Piauí, from January 2007 to March 2010. Standardization showed that 0.25 µg protein/well, a 1:200 dilution of the serum and concentration of 1:3,000 of the conjugated anti-goat IgG presented the best results. It was observed that the Agar Gel Immunodiffusion (AGID) detected 128 (18.4%) positive samples, and ELISA, 259 (37.2%). The sensitivity and specificity of i-ELISA regarding AGID were 94.5% and 75.7%, respectively. A higher prevalence was observed among animals older than six months (p < 0.05). The prevalence among males was of 56.7%, and among females, 35.4% (p < 0.01).
